As a new peptide-based drug moves from discovery to purification for clinical trials to commercial production, emphasis shifts from rapid, high performance synthesis and purification to improving the synthesis step to minimizing failure sequences and optimizing purification for yield and recovery. Agilent offers a full portfolio of HPLC columns, synthesis resins, acidic ion pair extraction tools and systems – and experience developed over many years serving the industry for customers to draw on.

Peptide solid phase synthesis solutions

* StratoSpheres resins ensure exceptional reproducibility and control of the resin linker load. They are available for Fmoc and Boc synthesis strategies and production of C-terminal peptide acids and peptide amides and are also available pre-loaded with the initial amino acid.
* AmphiSpheres are new amphipathic resins for a broader range of solvents and are an excellent alternative for difficult to synthesize peptides.

An array of purification media

* VariTide RPC columns and media are a 'universal' product for high performance purification of small quantities of synthetic peptides. It is a 6 µm material with a pore size optimized for peptides of 6 to 60 amino acids in length and using acidic, neutral and basic eluents.
* SepTech ST150 10-C18 - a silica material optimized for synthetic peptide purification using acid eluents.
* Polymeric reverse phase PLRP-S is the material of choice for GMP production (API purification) as it is manufactured in batch sizes ranging from 1 kg to 200 kg (600 L packed column) under a Type II Drug Master File. PLRP-S 100Å and PLRP-S 300Å columns and bulk media come in a range of particle sizes with options for high performance, high throughput and high yield purification. * For ion exchange purification, PL-SAX 1000Å, a strong anion exchanger and PL-SCX 1000Å, a strong cation exchanger are available pre-packed or in bulk.

Acid removal resins

* Post reverse phase purification is a finishing process that removes acidic ion pairing agents from the HPLC fraction and/or peptide. At the discovery stage, Agilent’s VariPure IPE acid removal resin is supplied in prep-packed SPE tubes.
* At development and production scales, PL-HCO MP resins are used to remove TFA (trifluoroacetic acid) and provide the peptide in the free-base form, and where acid labile, to minimize acid degradation and maximize yield.

Instrumentation for every stage

* For discovery, the 940-LC is a fully integrated, preconfigured, pretested system. A unique Scale-Up Module enables fast, easy transition from analytical to semi-preparative operation.
* At development, the PrepStar SD-1 system generates an accurate gradient at any flow from 0.01 to 200 ml/min without changing pump heads. Multi-gram level collection is from pre-packed columns and/or Load & Lock columns with high-resolution media.
* In production, the PrepStar SD-2 1200ml/min system is a high-throughput system ideally suited for purifying material for pre-clinical and early clinical trials. Load & Lock columns provide loading up to 20% greater than most DAC columns. For larger scale production skid systems and process HPLC columns are available.
