Calidifontibacter indicus gen. nov., sp. nov., a novel actinobacterium belonging to family Dermacoccaceae isolated from Western Ghats region, India

Calidifontibacter indicus gen. nov., sp. nov., a novel actinobacterium belonging
to family Dermacoccaceae isolated from Western Ghats region, India and emendation
of the family Dermacoccaceae.

During the course of the study of bacterial diversity of Western Ghats, India, an
actinobacterium strain designated as PC IW02(T) was isolated and characterized by
polyphasic taxonomic approach. The isolate PC IW02(T) was Gram positive, short
rod, non-motile and creamish in colour. The 16S rRNA gene sequence analysis
showed closest sequence similarity with members of the genus Dermacoccus:
Dermacoccus barathri (96.6%), Dermacoccus profundi (96.5%), Dermacoccus abyssi
(96.4%) and Dermacoccus nishinomyaensis (95.9%). The phylogenetic tree suggests
that the strain PC IW02(T) could represent a new genus of the family
Dermacoccaceae with the genus Demetria as closest clade. The pairwise sequence
alignment with the genus Demetria terragena HKI 0089(T) and Kytococcus
sedentarius DSM 20547(T) showed a similarity of 94.2% and 93.7% respectively. The
strain PC IW02(T) has MK-8(H4) as a major menquinone. C(16:0) ISO (43.4%) and
C(16:1) ISO H (17.2%) and C(17:0) anteiso (9.9%) were present as major fatty
acids. The diagnostic cell wall amino acid at position 3 of the peptide subunit
is lysine; the interpeptide bride consists of Gly-Ser-Asp. The polar lipids
present were diphosphatidylglycerol (DPG), phosphatidylglycerol (PG) and
phosphatidylinositol (PI) and phosphatidyl inositol mannosides (PIM) and
phosphatidylserine (PS) along with two unknown phospholipids. The genomic DNA G+C
contents of the strain is 77 mol%. On the basis of phenotypic characteristics
including chemotaxonomic data along with 16S rRNA gene sequence similarity the
strain PC IW02(T) should be considered as a novel genus of the family
Dermacoccaceae for which the name Calidifontibacter indicus gen. nov. sp. nov is
proposed. The type strain is PC IW02(T) (=MTCC 8338(T) =JCM 16038(T) =DSM
22967(T)). An emendation of the family Dermacoccaceae is provided.
